چکیده

Introduction This discussion will focus on two areas. The first will deal with the release and movement of iron from intracellular storage sites to the vascular compartment following an ischaemic episode. The conditions necessary for this release and the question of whether the quantities of iron involved are relevant to clinical situations occurring in acute-care medicine will also be addressed. The second area of discussion will focus on a number of preclinical studies involving the use of deferoxamine or a highmolecular mass analogue of deferoxamine in the treatment of a reversible myocardial dysfunction induced by short-term regional ischaemia. The critical role of ‘free’ iron in catalysing reactions leading to the formation of oxygenand lipid-derived reactive species has been reviewed extensively by several authors [ 1,2] and will not be discussed here.

Candesartan Attenuates Ischemic Brain Edema and Protects the Blood–Brain Barrier Integrity from Ischemia/Reperfusion Injury in Rats

Background: Angiotensin II (Ang II) has an important role on cerebral microcirculation however, its direct roles in terms of ischemic brain edema need to be clarified. This study evaluated the role of central Ang II by using candesartan, as an AT1 receptor blocker, in the brain edema formation and blood-brain barrier (BBB) disruption caused by ischemia/reperfusion (I/R) injuries in rat. Methods...

Role of iron chelation in hemorrhagic myocardial infarction: a quantitative CMR study

Background It has been speculated that iron chelation may be beneficial in acute myocardial infarction (AMI) and that early treatment can limit ischemia-reperfusion injury and also reduce infarct size. However, the role of iron chelation in hemorrhagic myocardial infarction, where it would be most suited, has not yet been explored. Reperfusion hemorrhage results in accumulation of iron degradat...

Prevention of postischaemic lipid peroxidation and liver cell injury by iron chelation.

Liver ischaemia was induced by cross clamping the hilar pedicle for 30 minutes in groups of rats with or without treatment with the iron chelating agent desferrioxamine (deferoxamine, DFR). The groups included eight animals each and were divided into the following treatment categories: control; ischaemia alone; ischaemia with subsequent reperfusion; ischaemia preceded by DFR, 60 mg/kg body weig...

Regulation of xanthine oxidoreductase by intracellular iron.

Xanthine oxidoreductase (XOR) may produce reactive oxygen species and play a role in ischemia-reperfusion injury. Because tissue iron levels increase after ischemia, and because XOR contains functionally critical iron-sulfur clusters, we studied the effects of intracellular iron on XOR expression. Ferric ammonium citrate and FeSO(4) elevated intracellular iron levels and increased XOR activity ...

Current state of clinical translation of cardioprotective agents for acute myocardial infarction.

There is continued interest in the concept of limiting myocardial infarct size with adjunctive agents administered along with reperfusion injury; however, there remains considerable controversy in the literature. The purpose of this article is to review the medical literature on clinical trials performed during the past 3 years that have attempted to reduce myocardial infarct size by administra...
